教育及工作經歷:
1982年吉林大學物理系,獲學士學位。
1982-1983年長春光機所光學專業,獲碩士學位。
1988年倫敦大學帝國理工學院物理系博士,專業方向為激光光譜、原子分子物理。
1988-1989英國倫敦大學帝國理工學院激光聯合體博士后。
1989-1991年大連化學物理研究所分子光譜實驗室博士后。
1991以后加入清華大學物理系原子分子納米科學教育部重點實驗室,任講師、1993年晉升副教授、2001年晉升教授、博士生導師。
2003年4月加入清華大學深圳研究生院創建“光學檢測與成像實驗室”。
2003.3.20-2014.1.20清華大學深圳研究院副院長。
2014.1.20至今,清華大學深圳研究院生命與健康學部主任。

生物光子學(2006年之前):包括:共焦與雙光子熒光成像、光學二次諧波成像、熒光各向異性度成像、熒光漲落譜,上述方法在生命科學研究中的應用。
生物醫學光學(現在):基于光散射的無損成像方法及其應用研究,包括:
光學相干斷層成像
偏振光散射理論、方法和應用,包括:偏振光散射理論、模型與模擬方法,偏振特征表征與提取方法,偏振敏感測量與成像方法,偏振光散射的應用。
研究方向:
1991-1997年以前從事專業方向為激光光譜技術和原子分子物理,1998年以后從事生物物理實驗方法研究,研究方向為顯微成像與微區熒光分析。2004年赴清華大學深圳研究生院創建“光學檢測與成像實驗室”,展開多項研究內容,包括:生物組織中偏振散射與傳播行為研究、偏振光成像方法與應用研究、非線性光學顯微成像方法與應用研究。
偏振光散射與應用研究:
偏振是光的基本特征之一,但也是多年來一直被忽視的一個特征。我們周邊無處不在的光或多或少都有一些偏振成分,所有的物體都會改變光的偏振。偏振光的狀態采用四分量斯托克斯(Stokes)向量描述,其中一個分量是光強。探測光的偏振態,可以用一個4x4穆勒(Mueller)矩陣描述所有物體的偏振光學性質。由于人眼對偏振不敏感,我們只能看到周邊世界光強變化,即穆勒矩陣16個陣元中的一個。因此,偏振光測量包含更加豐富的信息。并且偏振信息對亞波長微觀結構更加敏感。研究、發展偏振光散射測量方法,可以大大豐富我們對周邊世界的感知能力。

研究內容
偏振光散射理論與計算模擬:利用各種單次散射理論,研究不同散射模型下偏振光在復雜介質中散射傳播過程,特別是偏振指標與微觀結構的對應關系。研究內容包括:單次散射理論與方法,散射模型,散射仿體,CPU與GPU模擬方法。
偏振光測量:研究不同偏振量測量方法和誤差傳遞規律,包括:分時與同時穆勒矩陣測量方法與技術,偏振測量誤差傳遞,對特異性結構敏感偏振量的測量方法。
偏振信息提取與特異性指標:獲得特異性微觀結構對應的偏振指標,包括:穆勒矩陣分解與變換,偏振態顯示,大數據關聯與統計分析。
偏振光散射應用:研究偏振散射方法在固體微觀結構(生物組織、材料)、氣體顆粒物和海洋微生物測量中的應用。

光學檢測與成像實驗室
實驗室簡介:
光學檢測與成像實驗室于2004年5月成立,2010被評選為深圳市重點實驗室。實驗室的科研隊伍由四名教授、一名副教授、兩名講師、一名實驗員和三十多名博士、碩士研究生組成。實驗室在研究生院支持下,現擁有實驗儀器設備固定資產超過800 萬,包括熒光顯微鏡、多種半導體光源(包括寬帶高功率SLD 和不同波長的多個全固化連續激光器)和超快激光光源、光電倍增管和制冷CCD等探測器、數字示波器,以及各類光學、光機械元器件和光學檢測所需的一些基本科研裝備。驗實室面積近300 平方米,擁有光學平臺9個、以及光學暗室、光電子學實驗室、光學鍍膜實驗室以及機械加工等輔助設施,各類微機三十多臺。
招生信息:
目前實驗室每年招收理學碩士研究生5-7名;生物醫學工程碩士研究生5-7名;理學博士研究生3-5名;除此之外,還招收清華大學-伯克利學院博士研究生2名,碩士生2-5名。

主要成果
2004年實驗室成立以來已在國內外重要期刊上發表論文一百八十余篇,申請國家發明專利16項,美國專利1項。研制出適用于材料生產的在線監測、人工組織工程的在線監測、中醫形態學診斷等領域的光學檢測成像系統共6套,應用于臨床眼科診斷成像系統已經轉移到深圳市企業,產品已經推向市場。實驗室成員獲得2004年和2007年國家發明二等獎,2005年科學技術一等獎,2009年廣西科技進步二等獎一項。

馬輝:多個角度看世界
近年來,3D電影越來越受到人們的追捧。走進3D電影院,帶上一副偏振眼鏡,立體影像便展現在眼前。
其實,3D電影只是偏振應用的冰山一角。很少有人意識到,我們周邊實際上是一個偏振光的世界。通過對偏振光性質的測量,我們可以檢測區分不同的癌變組織,可以分辨不同粒徑和成分的氣體污染顆粒物,可以對海洋中的微生物進行現場計數與細致分類……而這些都是清華大學深圳研究生院教授馬輝正在研究的內容。
光學方法是使用時間最長、基礎積淀最深厚、應用范圍最廣泛,并且仍然迅速發展的觀測手段。“照圣經的說法,上帝創造世界做的第一件事就是創造光。”馬輝告訴記者,人類最早就是通過眼睛和光認識周邊世界,隨后用望遠鏡認識星空宇宙,用顯微鏡認識微觀世界,光學測量方法的發展伴隨著自然科學發展的整個歷程。
如今,光學仍然是一個非常具有活力的科學領域,并不斷獲得諾貝爾獎。例如,2008年的綠色熒光蛋白(GFP)、2009年的光纖和CCD以及去年的LED和超分辨成像。“如果你把這些成果串起來,你會發現在這短短幾年中,光學成像方法全鏈條各個環節都獲得了諾貝爾獎:GFP對樣品實現分子特異性對比度增強,LED是新型光源,光纖代表一種新型光路器件,CCD是新型探測器,而超分辨成像則是新的光學成像機理。”可以看出,古老的光學觀測方法正不斷煥發出新的生命力!2013年,聯合國確定2015年為“光之年”。
偏振是光的一個屬性,但很不幸,人類以及絕大多數動物的眼睛對光的偏振并不敏感。因此,盡管偏振這個詞大家都耳熟能詳,但偏振方法的潛力一直沒有得到真正重視,做偏振研究的人很少,偏振的應用也很少。從2004年開始,馬輝將偏振光散射這個看起來并不時髦的領域作為自己的研究方向。
十幾年來,人們在越來越多的領域開始意識到偏振測量的潛力,馬輝滿懷自信地說:“偏振可以大大增加光學方法獲取信息的能力,它將是光學發展的新方向,在信息技術突飛猛進的今天,偏振方法前景無限!”。
實現癌癥早期檢測
馬輝告訴記者,光學方法很適合于生物樣品。它基本不產生損傷,分辨率可以達到亞細胞層次,還可用于從活細胞到組織、器官和個體等不同層次的生物樣品。“但生物組織是不透明的,因為它有散射,當激光照射進人體,光能透進去,但看不清里面的血管和骨頭。散射破壞了光的有序性質,這是光學方法用于生物組織最大的問題。”馬輝解釋道,“利用偏振方法,可以抑制多次散射退偏光子對測量結果的貢獻,提高生物組織淺表層測量的分辨率。”
馬輝最開始研究偏振方法的一個基本目的就是降低散射的影響,提高成像質量。“人們發現,85%以上的癌變首先在上皮組織中產生,提高表皮組織成像的分辨率有可能幫助醫生發現早期癌變。”
偏振測量的另一個功能是獲得生物組織微觀結構的信息。馬輝告訴記者,散射可以改變光的方向,破壞成像結果,但散射也可以影響光的偏振,帶來新的信息。通過研究散射光偏振的變化與散射體的大小、密度、形狀和結構之間的關系,人們可以獲得樣品豐富的微觀結構信息,由此發現生物組織的異常和病變,實現病理診斷。
此外,偏振可以幫助提高淺表層組織成像的分辨率,還能夠測量生物組織微觀結構的變化,在癌癥早期診斷方面具有很好的應用前景。馬輝介紹,他們正與深圳南山醫院病理科合作測量癌變組織。“全偏振成像叫穆勒矩陣成像,我們從醫院找來很多不同類型的癌變樣品做穆勒矩陣成像,包括皮膚基底細胞癌、甲狀腺乳頭狀腺癌、乳腺癌以及宮頸癌等等。現在我們已經有一些證據,能證明全偏振成像的確有可能用于癌變的定量表征,但是我們還需要繼續搜集大量的樣本積累數據。”
2015年,馬輝獲得了國家重大科研儀器研制項目,他將利用這幾年的最新基礎研究成果和技術進展研發模塊化穆勒矩陣顯微鏡,演示其典型臨床應用。同時,他還將建立實驗室研究平臺,針對各個模塊開展原理和方法學探索和工程開發,研究信息提取方法和大數據分析方法,不斷改進儀器,推動產業化。
“2014年,超分辨成像的其中一個諾貝爾獎獲得者赫爾,首先在激光成像中實現了超分辨,但他采用的方法很復雜,不好使用。但赫爾很機靈,他讓萊卡公司把他的方法做成了儀器,給生物學家用。盡管這個儀器現在仍然要賣一千萬人民幣,并且仍然不那么好用,但由于生物學家展示了他的巨大應用價值,白茲格利用貝爾納的工作成果實現了另一種更好用、更便宜的光學超分辨成像方法,最后三個人一起得獎。”馬輝總結道,“光有好概念、好方法還不行,必須推廣出去,讓企業介入把方法變成產品,供更多的人使用。我這個儀器項目就是要發展一系列方法和裝置,并且拉上企業做儀器,為生物學家和醫生提供新的工具,提出新的需求,帶動偏振方法的研究。”
作為探索者,馬輝一路走來碰壁不少,他說:“走很多彎路之后最后發現碰對了,也是一種享受。我覺得做科研就要經歷這些過程!我會沿著這條路繼續往下走,把偏振的潛力不斷挖掘出來。”
偏振開啟一扇門
馬輝做偏振成像和應用是半路出家,他1988年在英國帝國理工學院獲得博士學位,1989年回國,后來加入清華物理系工作,有十幾年時間他一直從事原子分子光譜基礎研究。1995年對馬輝來說是一個轉折點。這一年,他在美國麻省理工學院訪問,與世界頂級科學家合作,研究雙原子分子高激發態光譜。
這一年中,馬輝參觀了很多實驗室。“我在麻省理工學院了解到利用拉曼光譜發現癌變和利用光學相干斷層成像測眼底的工作,在麻省綜合醫院參觀了交叉學科的威爾曼實驗室,在阿貢國家實驗室參觀了高分子光電轉換材料研究組,這些都給了我非常深刻的印象。”這些交叉學科前沿研究讓基礎研究出身的馬輝思索起來,“那一年我確實開始有些想法,覺得自己還是喜歡做偏應用一些的研究”。
回國后馬輝逐步把工作重點轉向生物物理實驗方法研究。從1998年到2003年,他率先在國內開展雙光子熒光成像和光學二次諧波成像,與國際上基本同步。他還建立了熒光漲落譜分析方法,并在熒光偏振成像方面開始探索,“我覺得那時候還真是進展很快,每天都有新的東西”。
2003年,馬輝被調到剛剛建立的清華大學深圳研究生院。深圳是中國改革開放的窗口,有突出的創新文化和快速成長的高技術產業,但高等教育卻滯后于經濟發展。“深圳有著一個很強、很完善的產業鏈結構,而且非常重視應用。在深圳做事,不得不考慮創新將來可能用在哪,能產生多大的影響。”脫離于北京裝備精良的實驗室,馬輝重新思索科研方向。“深圳研究生院有兩個特點,一個是產業合作,一個是學科交叉。既然來到深圳,就必須充分利用這些特點。”
2004年,從英國回來的一位老師加入了馬輝課題組。“他曾做過OCT方面的研究,于是我們決定做OCT產業化。為了同OCT的研究互相呼應,我也開始進行偏振光散射及其應用研究,這是一個全新的研究方向!”
馬輝向記者介紹,OCT就是光學相干斷層掃描儀的簡稱,它可以通過背向散射對眼底視網膜或眼角膜進行微米量級高分辨成像,為醫生提臨床診斷信息。課題組與當地公司合作,開啟了OCT產業化之路。
時至今日,當初合作的小公司已經占據中國眼科OCT產品30%以上市場份額,實驗室畢業的幾個碩士也成為了公司的骨干。與此同時,針對偏振光散射的研究也日益深入,從最初一個應用小課題,逐步產生新問題,帶出新模型、計算程序、實驗方法和應用,形成了完整的研究方向。回想整個發展歷程,馬輝十分感慨:“當初做偏振成像的時候我并沒有很高的期待值,周圍朋友也不認為它有多大的前途,但深入做下去就闖出一條路,而且可能是一條康莊大道。我們周圍是一個偏振光的世界,當你能感受偏振的時候,你會發現這個世界如此豐富多彩!”
最近,偏振研究越來越引起人們的關注,馬輝帶領團隊堅持了10余年,已經形成自己的研究特色,在國際上嶄露頭角。目前馬輝在偏振上專注于做四件事,“第一是測量,我們發展了一系列方法,能夠測得更加準確和迅速;第二是信息提取,就是針對不同應用尋找一些具有特異性的偏振指標,比如某種癌變的臨床診斷指標;第三是信息解釋,就是要知道不同微觀結構如何影響散射光偏振態變化,為解決逆問題提供支撐;第四就是應用,千方百計把偏振用于解決各種實際問題。應用也是我認為最重要的一件事”。
做一個牽線人
對馬輝來說,偏振能為我們提供一個信息寶藏。他打了一個形象的比喻,“不考慮偏振的時候,我們相當于用一個字母來給不同人命名,也就是只能用強度編碼。甲叫a,乙叫aa,丙叫aaa……而偏振能讓我們用單詞命名,jia、yi、bing……穆勒矩陣的16個陣元相當于給了我們16個字母,可以形成各種組合,我們描繪周邊世界的能力一下子提高很多”。另外,偏振方法具有普適性,現有光學方法都可能通過偏振調制升級為偏振方法,應用于不同類型的問題和樣品。
2013年,“霧霾”成為年度關鍵詞,世界上污染最嚴重的10個城市,有7個在中國。為此,我國在霧霾高發的16個省份選擇了43個監測點,以期掌握不同地區PM2.5污染特征及成分差異。“中國的污染很復雜,往往不知道污染源是什么,我們正在同一家公司和環保部門合作,發展一種利用偏振散射對空氣中顆粒物進行計數、分類和源解析的儀器”,他進一步解釋,“這就像一個農貿市場,原先我們只能估算人流總量,現在能告訴你不同時刻男人、女人、小孩各有多少,便于優化管理。”
幾年前,馬輝參與了國家海底觀測試驗網項目,負責數據管理系統。專業敏感性使馬輝開始思考如何將偏振應用到海洋,“通過這個項目我知道了很多海洋方面的需求,我們可以利用偏振方法升級現有的光學技術,提高通過測量所能獲得的信息量”。
從上世紀80、90年代開始,全球平均氣溫比100年前上升了0.48℃,科學家們針對全球變暖這一自然現象提出了種種猜測。馬輝告訴記者,“其實藻類對環境變化影響很大,通過監測海洋里不同類型的藻類,將不同海域里不同藻類的濃度等測出來,科學家就能探究氣候變暖的原因”。
馬輝常將自己的工作比喻為“造槍”,“我們就是要提供新型手段,幫助別人獲得信息。我就像一個造槍的,我不打仗,但我幫助打仗的人,提升軍隊的戰斗力”。
從事科學研究,興趣莫過于最好的助推劑。“我覺得發現問題之后解決問題很好玩,我就是喜歡做科研。”雖然時常奔波于全國各地甚至國外,但馬輝說:“這些年為科研而奔波,我還是挺享受的。”
在馬輝眼中,偏振好似一顆正在發芽的種子。數年來,在他的悉心呵護下,這顆種子破土而出,漸漸長大。他相信偏振在未來的應用潛力終將被更多人認知,就像彩色電視替代了黑白電視,偏振下的新景色將給人類帶來一個翻天覆地的變化。
